Yes, the strings do tighten the neck, but it shouldn't affect it too much. The only thing to look out for with changing the guage on the Mustang is to check that the tremolo tailpiece is still floating (perpendicular to the body). If it isn't then you may have troubles with it going out of tune. It can be fixed by adjusting the springs under the plate.
